Quality evaluation of Polyporus umbellatus based on HPLC specific chromatogram and fingerprint analysis

    Hong Wang is an associate professor of Department of Natural Medicine, School of Pharmacy, Peking University, Beijing. Her research interests include active components in medicinal plants and quality evaluation system of traditional Chinese medicine. She has undertaken many scientific research projects of Ministry of Education of the People's Republic of China, National Major Scientific and Technological Special Project for "Significant New Drugs Development" during the Twelfth Five-year Plan Period, National Administration of Traditional Chinese Medicine and Chinese Pharmacopoeia Commission. She also won the Second Prize of the State Scientific and Technological Progress Award and the third prize of Beijing Science and technology Progress Award. She made a systematic study on the resources and quality standards of many kinds of TCMs, including Rubiae Radix Et Rhizoma, Magnoliae Officinalis Cortex, Pyrrosiae Folium, Mume Flos, Polyporus and Apocyni veneti Folium. The results of her researches were presented in the Chinese Pharmacopoeia or in the industry standard and applied in practice. Professor Hong Wang has published more than 80 articles, wrote and edited 8 professional books. She is the editorial board member of Chinese Journal of Experimental Traditional Medical Formulae, the peer review expert of Beijing Natural Science Foundation, and the member of council of the Medicinal Resources Branch of the China Medical Association of Minorities.

摘要:

本研究基于高效液相色谱法(HPLC)建立了一种简便、可靠、重现性好的中药猪苓特征图谱和指纹图谱分析方法, 对不同来源的猪苓药材及饮片样品进行分析。最终, 在特征图谱中共确定了10个特征峰, 并将其应用于猪苓药材及饮片的真伪鉴定; 在指纹图谱中共确定了9个共有峰, 并对32批猪苓药材及饮片样品进行相似度计算。采用HPLC-APCI-IT-TOF-MSn法对猪苓的主要成分进行鉴定, 共鉴定了8个化合物, 其中在特征图谱中指认了4个色谱峰, 在指纹图谱中指认了4个色谱峰。本研究首次将HPLC特征图谱与指纹图谱相结合, 应用于猪苓真伪鉴别和质量评价, 为建立系统完善的猪苓质量标准提供了新方法。

Abstract:

To evaluate the quality of Polyporus umbellatus, we established a simple, repeatable and reliable method based on high performance liquid chromatography (HPLC) for specific chromatogram and fingerprint analysis, which was applied to analyze samples of medicinal materials and decoction pieces collected from different regions. Finally, ten characteristic peaks were designated in the specific chromatograms and applied to the authenticate identification of P. umbellatus samples. Nine common peaks were designated in the fingerprints, and then the similarities between 32 batches of samples were calculated. Among them, eight compounds were identified by HPLC-APCI-IT-TOF-MSn, four of which were identified in specific chromatograms and four in fingerprints. In the present study, we, for the first time, combined HPLC specific chromatograms and fingerprints for the species identification and quality evaluation of P. umbellatus. Collectively, our findings provided a new method for establishing a comprehensive quality standard of P. umbellatus.
