HPLC-PDA 测定黄桷树叶提取液中绿原酸和芦丁的含量

Determination of chlorogenic acid and rutin in extractive solution of ficus virens leaves by HPLC-PDA

摘要:

建立测定黄桷树叶提取物中绿原酸和芦丁含量的HPLC-PDA法, 采用ODS2色谱柱, 梯度洗脱, 流动相甲醇–磷酸溶液(pH 3.0), 柱温25 ± 2 oC, 流速1.0 mL/min。通过质谱和紫外光谱技术对提取物中有效成分进行结构确证。经确证提取物中含有绿原酸和芦丁, 所建立的测定绿原酸和芦丁含量的HPLC-PDA法, 在所考察的浓度范围内线性关系良好, r ≥ 0.9998。定量限均小于0.06 μg/mL, 检测限均小于0.02 μg/mL; 相对回收率范围为99.10%–104.7%。以RSD值表示的日内精密度均小于1.89%, 日间精密度均小于1.82%。该方法灵敏度高, 准确度好, 专属性强, 操作简便, 可用于黄桷树叶提取液中绿原酸和芦丁的含量测定。经测定黄桷树叶提取液中芦丁和绿原酸含量分别为3.46和3.23 μg/mL。

Abstract:

A HPLC-PDA method was established for determining chlorogenic acid (CGA) and rutin (RU) in the extract of Pueraria lobata leaves. The ODS2 column was used with gradient elution, the mobile phase was methanol–phosphoric acid (AR) solution (pH 3.0), the column temperature was 25 ± 2 oC, and the flow rate was 1.0 mL/min. The effective components in the extract were identified by mass spectrometry and UV spectrometry. The results showed that the extraction contained CGA and RU. The HPLC-PDA method for determining CGA and RU showed an excellent linear relationship within the concentration range, r ≥ 0.9998. The results showed that the limits of quantitation and detection were all less than 0.06 and 0.02 μg/mL, respectively. The relative recoveries ranged from 99.10% to 104.7%. The intra-day and inter-day precisions expressed by RSD values were all less than 1.89% and 1.82%, respectively. The method was sensitive, specific, and easy to operate and could be used to determine CGA and RU in the extract of Pueraria lobata leaves. The contents of RU and CGA were 3.46 and 3.23 μg/mL, respectively.
