中药玄参中哈巴俄苷和肉桂酸的高效液相色谱法测定

HPLC Determination of Harpagoside and Cinnamic Acid in Radix Scrophulariae

摘要: 目的: 建立中药玄参中哈巴俄苷和肉桂酸的高效液相色谱含量测定方法, 提出特征性有效成分哈巴俄苷的含量限度建议。方法: Ultrasphere ODS 分析柱(250 mm×4.6 mm, 5 mm), 流动相为乙腈和1% 醋酸水溶液, 线性梯度洗脱(20:80→50:50) (20 min), 流速为1 mL·min-1, 室温, 检测波长为278 nm结果: 哈巴俄苷与肉桂酸回收率分别为97.13% 99.38%, RSD分别为0.80% 0.51%, 全国20份商品饮片和8份不同产地完整药材中哈巴俄苷和肉桂酸的含量分别为0.041~0.244% 0.012~0.068%结论: 本文方法快速、简便、准确, 可用于玄参的质量控制。建议规定玄参中哈巴俄苷含量应不低于0.05%

Abstract: A gradient HPLC method was established for the determination of harpagoside and cinnamic acid in Radix Scrophulariae (Xuanshen) and a proposition was put forward for the lowest content of the characteristic and active constituent (harpagoside) for qualified Radix Scrophulariae. The experimental conditions were as follows: Ultrasphere ODS column (250 mm×4.6 mm, 5 mm), mobile phase: acetonitrile-water (containing 1.0% acetic acid) (20:80→50:50) (20 min), flow-rate 1 mL·min-1, room temperature, detection wavelength 278 nm. Twenty-eight samples of Xuan-shen (Radix Scrophulariae) from different districts of China were analyzed and the contents of harpagoside and cinnamic acid in Xuan-shen were 0.041~0.244% and 0.012~0.068% respectively. The recoveries (±RSD)% were 97.13(±0.80)% for harpagoside and 99.38(±0.51)% for cinnamic acid. The method is simple and accurate. It can be used for the quality control of Radix Scrophulariae. We propose that the content of harpagoside in qualified Radix Scrophularia should be no less than 0.05%.
