当归主要成分的液质联用鉴定及不同药用部分的比较

Characterization of principal compositions in the roots of Angelica sinensis by HPLC-ESI-MSn and chemical comparison of its different parts

摘要: 应用HPLC-ESI-MSn方法分析当归主要成分, 根据紫外和质谱特征指认了24个成分, 包括11个酚类、12个苯酞类和1个氨基酸, 其中3个二咖啡酰奎宁酸、5-阿魏酰奎宁酸和阿魏酸己糖苷为当归中首次报道。中医认为当归不同药用部分 (归头、归身和归尾)具有不同的功效, 但其药效物质仍不明确, 本文采用HPLC法对归头、归身和归尾的化学成分进行了比较, 结果从归头到归尾色氨酸的含量逐渐降低, 而苯酞类等低极性成分含量依次升高, 研究结果为进一步解释当归不同药用部分具有不同的功效提供了依据。

Abstract:

Angelicae Sinensis Radix, Danggui in Chinese, is an important traditional Chinese medicine derived from the roots of Angelica sinensis (Oliv.) Diels. HPLC-ESI-MSn was applied to analyze the chemical constituents in the methanol extract of Danggui. A total of 24 compounds were identified or tentatively characterized based on their UV and mass spectra, containing 11 phenolic compounds, 12 phthalides and 1 amino acid. Three dicaffeoylquinic acids, 5-feruoylquinic acid and ferulic acid hexoside were detected in A. sinensis for the first time. According to Chinese medicine theories, the three parts of Danggui (Guitou, Guishen, Guiwei) have different therapeutic effects and their substantial evidence has not been clearly elucidated yet. In the present study, the chemical compositions of different parts of Danggui were investigated by using the established HPLC method. The result indicated that the content of tryptophan was getting lower and the contents of low polarity constituents like phthalides trended to get higher from Guitou to Guiwei. This might partly explain the therapeutic effects of different parts of Danggui.
