聚(甲基丙烯酸-co-泊洛沙姆)水凝胶的溶胀和药物释放性质

Swelling and Drug Release Characteristics of Poly (methacrylic acid-co-poloxamer) hydrogels

摘要: 以自由基溶液聚合方法制备了聚(甲基丙烯酸-co-泊洛沙姆)共聚物水凝胶, 研究凝胶的溶胀和模型药物氢溴酸右美沙芬(DMP)和维生素B12的释放性质. 凝胶的溶胀和药物释放具有pH-依赖的特征, 在中性或碱性介质中的溶胀和药物释放速率要高于酸性介质中. 在中性或碱性介质中, 凝胶的溶胀为非Fick或零级过程, 对于组成适当的水凝胶, 在数小时内以零级动力学过程释放药物.

Abstract: Poly (methacrylic acid co-poloxamer) hydrogel networks were synthesized by free-radical solution polyme-rization, and the dynamic swelling and in vitro release properties of model drugs, dextromethorphan hydrobromide (DMP) and vitamin B12 (VB12) were studied. These gels exhibited pH-dependant swelling and sustained drug release properties, and the water uptake rate and drug release rate in neutral or basic media were higher than that in acidic media. The results showed that the water uptake followed non-Fickian or zero order process in neutral or basic media, and the release of model drugs from hydrogels of appropriate composition was of zero order kinetics over a period of several hours.
