槲寄生抗血小板激活因子活性成分的研究

A Novel Platelet-Activating Factor Antagonist Isolated from a Chinese Herbal Drug Viscum coloratum

摘要: 本文以血小板激活因子(PAF)和二磷酸腺苷(ADP)诱导的血小板聚集试验为检测指标, 追踪观测了中草药槲寄生对PAFADP诱导的血小板聚集的影响。从槲寄生乙酸乙酯提取物中分离得到的化合物高圣草素7-O-β-D-葡萄糖苷, 体外试验证实对PAF诱导的血小板聚集有明显的抑制作用, 而对ADP诱导的血小板聚集无显著性抑制作用。提示高圣草素7-O-β-D-葡萄糖苷是槲寄生抗PAF的有效活性成分。

Abstract: A novel natural platelet-activating factor (PAF) antagonist was isolated from a Chinese medicinal. herb Viscum coloratum and its structure was identified as ho moeriodictyol-7-O-β-D-glycoside, which existed in ethyl acetate extract fraction. At the final concentration of 10-5 mol·L-1, it could significantly inhibit human and rabbit platelet aggregation induced by PAF, with IC50 being 8.0×10-7 mol·L-1, but had no inhibitory effect on platelet aggregation induced by ADP.
