雷公藤中的一种新活性成分-16-羟基雷公藤内酯醇

16-Hydroxytriptolide: An Active Compound from Tripterygium wilfordii

摘要: 从雷公藤Tripterygiurn wilfordii Hook f. 的叶、根及制剂雷公藤多甙中分离出一个新的三环氧二萜内酯化合物-16-羟基雷公藤内酯醇L2。该化合物是一种白色簇状结晶, 熔点232~233.5 ºC, 分子式为C20H24O7。根据光谱UV, IR, MS, 1H NMR, 13C NMR, 2D NMRSelective Long-range DEPT数据分析, 确定了其化学结构, 并通过X-射线晶体分析确证了它的立体化学。15-C“S” 构型的手性碳。药理试验表明, 16-羟基雷公藤内酯醇具有较强的抗炎、免疫抑制和雄性抗生育活性。另外, 还分离到已知化合物雷公藤内酯醇, 并通过2D NMR等光谱分析归属了全部碳和质子的NMR光谱峰。

Abstract: From the dried roots and leaves of Tripterygium wilfordii 1.2, a new diterpenoid triepoxide, 16-hydroxytriptolide was isolated, and its structure and stereochemistry elucidated as 16-S-hydroxy-triptolide on the basis of spectral data IR, MS, UV, 1H NMR, 13C NMR, 2D NMR, Selective Long-range DEPT and x-ray crystallographic analysis. This compound showed definite antiinflammatory action, strong immunosuppressive and antifertile activities. In addition, a known compound, triptolide was also isolated and all the spectral signals of 1H NMR and 13C NMR were assigned.
