    The Effects of Ethanolamine Nitrate Ester Ferulate (ENF) on Isolated Working Guinea Pig Heart and Rat

    The cardiac and vascular effect of ethanolamine nitrate ester ferulate (ENF) wasinvestigated on isolated working guinea pig heart (IWH) and rat aortic ring. The coronary blood flowwas remarkably increased while the cardiac functions of IWH were not significantly altered by ENFranging from 10-8 to 10-4 mol/L. The percentages of vasorelaxation of ENF (10 μmol/L) and nicoran-dil (NIC, 10 μmol/L) on K+ (80 mmol/L)-induced contraction in aortic rings were 72±8% and17±8%, respectively. The relaxation effects of ENF and NIC on phenylephrine (PE, 5 μmol/L) in-duced contraction were in a dose-dependent manner. When vessel segments were exposed to PE withmethylene blue (MB, 10 μmol/L) or with glibenclamide (GLI, 10 μmol/L), the percentages of relaxa-tion of ENF (10 μmol/L) and NIC ( 10 μmol/L) were 41±11% and 49±7% or 76±14% and the 33±9%, respectively. The results suggested that ENF improved coronary circulation and the vasodilation of ENF was mediated bv nitrate-like action.

    The effect of Rg1, a saponin extracted froin Panax ginseng, on the phenotype, receptor and the activity of protein tyrosine kinase (PTK) of lymphocytes isolated from 7 healthy oldpersons were studied. The CD25, CD45RA and CD45RO phenotypes of lymphocytes were 4eter-mined by indirect immunofluorescence technique. The percentage of CD25, CD45RA and CD45RO positive lymphocytes was 38.3%±17.3%, 46.0%±15.1%, and 52.6%±14.1% respectively after incu-bation with PHA (5 μg/ml) for 72 hours. However, there were 58.0%±12.5%, CD25, 64.1% ± 12.4%, CD45RA, and 74.0%±8.0%, CD45RO positive cells in the presence of Rg, ( 1μg/ml) along with PHA(5 μg/ml) over the sanie period of incubation. A significant increase was induced by Rgi (P<0.05).The activities of PTK in the cytoplasm and membrane of lymphocytes were measured by ELISAmcthod after incubation with PHA or PHA+Rg1. The absorbance value of PTK activity in cytoplasm after 72 h incubation was 0.120±0.020 in PHA group, but 0.138±0.015 in PHA+Rg1 group. In thelymphocyte membrane, it was 0.374± 0.060 in PHA group and 0.403 ± 0.008 in PHA+Rg1 group(P<0.001). These results showed that Rgi significantly arid simultaneously increased both the PT Kactivity and the expression of phenotype of lymphocytes.

    Ebselen. 2-phenyl-1,2-benzisoselenazol-3 (2H)-one, activated by. glutathione(GSH) was shown to react with N-acetyl-p-benzoquinone imine (NAPQI). the toxic rnetabolite ofparacetamol, but not to react with paracetamol. The GSH-activaled ebselen reacts uith NAPQI fasterand more complete than with ebselen alone. Compared xvith the UV-spectra of the solution containing20 μmol/L ebselen tactivated by 20 μmol/L GSH) and 20 μmol/L NAPQI with the solution contain-ing 20 μmol/L ebselen tactivated by 20 μmol/LGSH) and 20 μmol/L paracetamol, NAPQI was foundto be reduced to paracetamol by activated ebselen. The optimal PH range for this reaction was foundto be 7.4 to 8.2.

    In recent years, there have been dramatic increases in the production of North American ginseng around the world. These increases and the expectation of their continuation have given rise to a need to critically evaluate all aspects of North American ginseng (Panar quinquefolium L.) production. Employing a system approach, the production of North American ginseng can be divided into its principal components that include seed preparation and planting, growing environment creation and maintenance, plant health maintenance, harvest and post-harvest activities, and product processing. As competitive industry forces are beginning to result in increased global competition, it is prudent to carefully assess both the strengths and weaknesses of the North American ginseng production system. It is clear that the utilization of appropriate technology will be a key to future industry successes and the continuity of successful economic development that is associated with the industry. It is recognized that careful technological choices and changes will be required to successfully face the challenges of an ever increasingly competitive global ginseng marketplace.

    The pharmacokinetic-pharmacodynamics of propranolol (PPL) and its active metabolite 4-hydroxypropranolol (4-OH-P) was studied on Chinese subjects by single or multiple oral administration. The efficiency of beta-blockade was measured as the reduction of heart rates or blood pressure in the supine and up right positions during rest or exercise period. After a single dose of 40 mg PPL, the plasma concentration of 4-OH-P was quite high, C(m)max and AUC (m) were 26.1±13.2 ng/ml and 180±69 ng.h/ml respectively, which were 50% and 73% of those of PPL. While after multiple dose administration, the plasma PPL concentration increased much greater than that in single administration and the 4-OH-P/PPL plasma level ratio fell from 0.79±0.64 at single dose to only 0.48±0.32 at steady-state. The pharmacodynamic half-life of PPL on inbibiting exercising heart-rate was much longer than the half life of drug concentration (8.78±2.27 vs 4.23±1.33 h. P<0.01).The Css 50. plasma concentration at steady-state producing 50% maximal efficacy, was 44.66±35.24ng/ml. The study showed that 4-OH-P is an important active metabolite of PPL and one of the possible factors causing the considering variations in the response to PPL in Chinese people.
